I have a character who has big ears like a donkey or mule, but has the build of a horse. Would it be possible for her to be a cross between a horse and mule? She is too horse like to be a mule herself

No, Mules cannot breed, any hybrid animal (Mules, Zebroid, Ligers and any other such animals) can't breed.

I think you mean "Can a Donkey and Horse breed" Donkeys and Horses can breed/mate and produce what is known as a Mule.
